Intestinal epithelial cell mucin glycoproteins—chiefly Mucin 2 (MUC2)—are large, heavily O-glycosylated proteins secreted primarily by goblet cells in the intestinal tract. Mucins form the mucus layer, a dynamic gel that constitutes a physical and chemical barrier protecting the epithelial lining from pathogens and commensal microbes, mediating immune responses, and helping maintain gut homeostasis. The mucus barrier is organized into a dense inner layer, largely sterile and attached to the epithelium, and a looser outer layer colonized by bacteria. Disruptions in mucin production or structure are implicated in gastrointestinal diseases such as inflammatory bowel disease, increased susceptibility to infection, and disturbance of the gut microbiota. Mucins are not classical drug targets, but their regulation is central to intestinal health and disease pathogenesis.
